AB An investigation into the characterisation of mammalian nuclear proteins
which bind to the HIV-1 LTR U3 region has been carried out using a
series of overlapping 40-mer oligonucleotides. These oligonucleotides
were synthesised to correspond to the HXB2 sequence from n.t. -210 to
n.t. -454. Two oligonucleotides have shown a number of specific binding
interactions in mobility shift assays, U.V. cross-linked mobility shift
assays and south-western analysis. Oligonucleotide 7 (n.t. -210 to n.t.
-250) has at least 4 specific interactions and oligonucleotide 13(n.t.
-410 to n.t. 454) has at least 2 specific interactions. The region of
oligonucleotide 7 is thought to bind ILF (Li et al, 1991) and possibly
members of the ets family. No protein binding has been reported in the
region of oligonucleotide 13. Reference: Li P, et al. 1991 P.N.A.S. USA
88; 7739-7743.
